Workpackage 4 – Dynamics of chromatin-related proteins

Studying dynamics of chromatin-related proteins in living cells after induced DNA damage. Particularly to study dynamics of such proteins: HP1 (?, ?, ?), Jmjd2b (histone demetylase), BMI-1 (PcG protein).

Description of work

Dynamics of of selected proteins will be studied in irradiated cells. We will use mouse SUV39h wt and dn cells in order to analyze the influence of changes in histone code on DNA repair mechanisms. For such analysis we will use highresolution confocal microscopy and UV lasers 355 nm and 405 nm. Induction of DNA strand breaks will be recognized by antibody against phosphorylated H2BX. We are going to find if
selected chromatin-related proteins are involved in DNA-repair machinery.
